發展高生物可利用率與低毒性岩藻糖報告子應用於醣基化細胞影像與生物標記 Development of High-Bioavailability and Low-Toxicity Fucose Chemical Reporters for Glycan Imaging and Biomarker Discovery
Sialylation and fucosylation play critical roles in various cellular physiological processes, and their abnormal expression is closely linked to cancer. This project aims to explore the relationship between cancer and aberrant glycosylation by synthesizing novel fucose analogs that serve as highly sensitive and low-toxicity chemical reporters. Our approach involves using bioorthogonal chemistry to conjugate fucose reporters with fluorescent dyes in HeLa cells, enabling the visualization of glycosylated biomolecules through confocal microscopy. Furthermore, by integrating this cell imaging system with mass spectrometry, the project seeks to identify aberrantly fucosylated proteins. Ultimately, this research aims to discover cancer cell biomarkers and develop targeted therapies for cancer.
